The Effects of Various Agents on the Mast Cell in Vitro

Abstract
These studies were performed to observe the influence of colchicine, microtubule and microfilament disrupting agent, and antihistamine on the isolated rat peritoneal mast cells in vitro.
To the medium chamber containing 0.3 ml of the isolated mast cell suspensions pretreated with colchicine(10¢¥5M), 0. 03 ml of antihistamine (1.25mg/ml) was introduced respectively. And then the time-lapse cellular changes or degranulating response of the mast cell were examined by inverted microscope.
The following concluions were obtained.
Colchicine alone did not cause degranulation with the exception of various morphological changes of the mast cells Whereas antihistamine induced massive degranulation of mast cells. Such a degranulating respones of mast cells to antihistamine was decreased to 87% by pretreatment with colchicine. These results suggest that the action of antihistamine on the mast cell degranulation is related to the microtubule and microfilament within the mast cell cytoplasm.
